We are re-submitting our revised manuscript entitled "Collagen reorganization at the tumor-stromal interface facilitates local invasion" by Paolo P. Provenzano, Kevin W. Eliceiri, Jay M. Campbell, David R. Inman, John G. White, Patricia J. Keely for possible publication as an article in BMC Cancer. We have carefully addressed each of the reviewers' concerns and submit a point-by-point response to each of the reviewers' comments. Additionally, major changes to the text of the manuscript are highlighted with red font.

Collagen reorganization at the tumor-stromal interface facilitates local invasion

BACKGROUND Stromal-epithelial interactions are of particular significance in breast tissue as misregulation of these interactions can promote tumorigenesis and invasion. Moreover, collagen-dense breast tissue increases the risk of breast carcinoma, although the relationship between collagen density and tumorigenesis is not well understood. As little is known about epithelial-stromal interaction...

Contact guidance mediated three-dimensional cell migration is regulated by Rho/ROCK-dependent matrix reorganization.

Cells generate mechanical force to organize the extracellular matrix (ECM) and drive important developmental and reparative processes. Likewise, tumor cells invading into three-dimensional (3D) matrices remodel the ECM microenvironment. Importantly, we previously reported a distinct radial reorganization of the collagen matrix surrounding tumors that facilitates local invasion. Here we describe...

Histochemical Analysis of Collagen Reorganization at the Tumor-Stromal Interface in Oral Squamous Cell Carcinoma- A Polarizing Microscopic Study

Stromal changes are the key factors for provision of nutrition and growth to any tumor and also they act as a barrier for spread of the tumor. There are limited histochemical studies in the literature on methods to detect, quantify and analyze the collagen in the invasive tumor front (ITF) of oral squamous cell carcinoma (OSCC). Aim: 1) To analyze the nature of collagen (thickness, hue, density...
